Figure1.Western blot analysis of extracts of various lane1 Mouse brain(+++)、lane2  Rat brain(+++), using TUBB3 antibody (CM01147HuM10-C1F) at 2ug/ml Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) at 1:20000 dilution. Lysates/proteins: 25ug per lane.Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit . Exposure time: 1.5min.











 


Figure2.Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of mouse brain sections labelling TUBB3 with purified CM01147HuM10-C1F at 10ug/ml dilution . Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using citrate buffer (PH6.0). Tissue was counterstained with Hematoxylin. Mouse specific IHC polymer detection kit HRP/DAB secondary antibody was used at 1:2000 dilution. PBS instead of the primary antibody was used as the negative control.

背景信息There are five tubulins in human cells: alpha, beta, gamma, delta, and epsilon. Tubulins are conserved across species. They form heterodimers, which multimerize to form a microtubule filament. An alpha and beta tubulin heterodimer is the basic structural unit of microtubules. The heterodimer does not come apart, once formed. The alpha and beta tubulins, which are each about 55 kDa MW, are homologous but not identical. Alpha, beta, and gamma tubulins have all been used as loading controls. Tubulin expression may vary according to resistance to antimicrobial and antimitotic drugs.
