Figure1.Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human tonsil sections labelling MLKL with purified  PA00139HuA10 at 10ug/ml. Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using Bond™Epitope Retrieval Solution  (EDTA, pH 9.0) . Tissue was counterstained with Hematoxylin.Rabbit specific IHC polymer detection kit HRP/DAB secondary antibody was used at 1/4000 dilution. PBS instead of the primary antibody was used as the negative control.

背景信息Mixed lineage kinase domain like pseudokinase (MLKL), belongs to the protein kinase superfamily, has two MW of 54 and 30 kDa. MLKL plays a critical role in tumor necrosis factor (TNF)-induced necroptosis, a programmed cell death process, via interaction with receptor-interacting protein 3 (RIP3), which is a key signaling molecule in necroptosis pathway. High levels of this protein and RIP3 are associated with inflammatory bowel disease in children.
