Figure1.Western blot analysis of extracts of various lane1 mouse brain(+++)、lane2 Rat brain(+++), using MAP1LC3A antibody (MA01074HuM10-B1F) at 5ug/ml Secondary antibody: HRP Goat Anti-Rabbit IgG (H+L) at 1:20000 dilution. Lysates/proteins: 25ug per lane.Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit . Exposure time: 1min.











 


Figure2.Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of mouse brain sections labelling MAP1LC3A with purified MA01074HuM10-B1F at 30ug/ml dilution . Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using citrate buffer (PH6.0). Tissue was counterstained with Hematoxylin. Mouse specific IHC polymer detection kit HRP/DAB secondary antibody was used at 1:2000 dilution. PBS instead of the primary antibody was used as the negative control.

背景信息LC3A. Microtubule-associated proteins 1A/1B light chain 3A (also known as MAP1LC3A) is an Ubiquitin-like modifier of the ATG8 family and is involved in the formation of the autophagosomes. This protein interacts directly with Sequestosome-1 and is recruited to inclusion bodies containing polyubiquitinated protein aggregates where it plays a role in inclusion body degradation by autophagy. MAP1LC3A-Rhodamine110 (Rh110) is a fluorogenic substrate for the autophagy hydrolase ATG4B. This substrate is useful for studying ATG4B when detection sensitivity or continuous monitoring of activity is essential.
